Today, we discussed about our schedule for cell culture for the next month. Our goal is to compare Bone Marrow MSC in growth medium with differentiating Bone Marrow MSC. It usually takes 21 days for MSC to differentiate, so for us, we will check after 15 days to compare the differentiation at different time point.

Today, we changed the growth medium of the 2 dish of differentiating cells and 2 dish of cells in normal growth medium.

Next week, I will fix plate A and C, and change the medium of the others.

On March 12, I will fix B and D, and stain all of four plates.

A and B is our control group. The first stain I will be using is ALP (Alkaline Phosphatese ), and this will produce pink spot in the Osteo containing cell. The second cell I will be using is Von Kossa (silver nitrate solution), and this will produced darker black spot in the Osteo containing cell.
